Rat Strain Models for Cardiovascular Research

Hypertension Models

SS/JrHsdMcwi

This is a hypertensive strain which has many features of salt-sensitive hypertension in humans. These are salt-sensitive, hyperlipidemic, insulin-resistant and have a low-renin form of hypertension. Renal, cardiac and vascular end organ damage is observed after severe hypertension. (REF RGD ID: 727992)
Female-specific linkage analysis in 99 female F2 derived from male SS/JrHsdMcwi and female BN/NHsdMcwi where 126 QTL were identified.
Other Phenotypes…increased blood pressure (REF RGD ID: 628478), increased heart rate (REF RGD ID: 727992), increased kidney weight (REF RGD ID: 727992)

Sr/JrHsd

These are resistant to hypertension with a high salt diet. Generally used as a control strain for SS rats (REF RGD ID: 61053)

SS/Jr

In the presence of dietary salt as an environmental factor develops hypertension with age. This parental strain is used in 157 out of 420 QTL (REF RGD ID: 629622)

BN/NHsdMcwi

LH/Mav

These have increased body weight, hyperlipidemia and high blood pressure which are common features to the human metabolic syndrome.(REF RGD ID: 1302893). Heart rate is similar to LN/Mav at 5 wks of age. (REF RGD ID: 1580397)

SS-13BN/Mcwi

These consomic rats are a potential model for multiple renal diseases, myocardial infraction and hypertension. When fed a low sodium diet their aortas relaxed less than those of SS parental rats. (REF RGD ID: 2293556). Male rats have reduced proteinuria and albuminuria, when fed 8% NaCl diet mean arterial blood pressure was decreased (REF RGD ID: 2300225)

GH

A genetically inbred model for hypertension, these have an early onset of hypertension, hypercholesterolemia, cardiac hypertrophy, and vascular disease at the age of 6 weeks (REF RGD ID: 2301936). Their mean arterial blood pressure is significantly higher than BN. (REF RGD ID: 2301936).

LEW

Lewis rats are highly resistant to salt-induced hypertension, even though their blood pressure is lower than SR rats. These tend to loose weight when on 8% NaCl diet (REF RGD ID: 61040). Mutation in one of the Cyp4a gene may play a role in the hypertension observed in this strain (REF RGD ID: 1578407).

SHR

Spontaneously hypertensive rats are a great model for human insulin resistance syndrome as these have reduced insulin-mediated glucose disposal, defective metabolism of fatty acid (REF RGD ID: 619680). 

WKY

This is a normatensive strain having lower SBP and DBP as compared to SHR (REF RGD ID: 731210). These respond to stress hormonally and behaviorally as they have high serum thyroid-stimulating hormone (REF RGD ID: 1302891).

Aerobic Running Capacity Models

COP/OlaHsd

Male COP rats had the lowest treadmill running capacity; ran to exhaustion (262 meters); Male DA were not significantly different in body weight as compared to COP. Have significantly lesser heart weight and relative heart weight compared to DA. Used as a control for DA/OlaHsd. (REF RGD ID:619535). This strain is used in 3 out of 420 rat QTLs.

DA/OlaHsd

Male DA rats had the highest treadmill running capacity; ran to exhaustion (969 meters); Male DA were not significantly different in body weight as compared to COP. Have significantly greater heart weight and relative heart weight and 50% greater cardiac output as compared to COP. This parental strain is used in 5 out of 420 QTL.

Transgenic Models

SHRSP-Tg(Tagln-ACE2)6918Bdr

Expression of  human ACE2 leads to increased circulating levels of angiotensin resulting in reduced mean arterial blood pressure (REF RGD ID: 2302131). These have attenuated hypertrophy compared to SHRSP who develop cardiac hypertrophy at an early age (REF RGD ID: 2302131).
Other Phenotypes: cardiac fibrosis (REF RGD ID: 2302131), decreased mean arterial blood pressure (REF RGD ID: 2302131). 

F344-Tg(Cyp1a1-Ren2)10

These are a model of malignant hypertension. (REF RGD ID: 2302144). Transgene expression is dependent on the dose of xenobiotic indole-3 carbinol. This model shows a dose dependent and reversible expression of transgene and hypertensive vascular damage (REF RGD ID: 2302140).
Other Phenotypes: increased circulating renin level (REF RGD ID: 2302140), increased systolic blood pressure (REF RGD ID: 2302140), weight loss (REF RGD ID: 2302140).

SD-Tg(Ren2)27

These are a good model for low-renin hypertension, with high blood pressure leading to cardiovascular diseases and finally die due to cerebral haemorrhage (REF RGD ID: 628589).
Other Phenotypes: increased blood pressure (REF RGDID: 628589).

SHR-Tg(Ef1a-Cd36)10Ipcv

These have 6-8 copies of Cd36 genes from the wild type Wistar  which shows improvement in glucose tolerance, serum fatty acid levels and insulin-stimulated glucose tolerance (REF RGDID: 68930).
Other Phenotypes: decreased fatty acid level (REF RGDID: 68930), improved glucose tolerance (REF RGDID: 68930), insulin resisitance (REF RGDID: 68930).

SHR-Tg(Ef1a-Cd36)19Ipcv

These have a single copy of Cd36 genes from the wild type Wistar  which shows improvement in glucose tolerance, serum fatty acid levels and insulin-stimulated glucose tolerance. (REF RGDID: 68930).

SHR-Tg(Ef1a-Cd36)106Ipcv

These express wild type CD36 gene from Wistar which ameliorates insulin resistance, glucose tolerance and improves dyslipidemia (REF RGDID: 1600636).
