This genomic element is located in an intron of the Vav2 (vav 2 oncogene) gene on chromosome 2. It was predicted to be a cis-regulatory module (preCRM) based on the presence of non-conserved consensus binding motifs for the essential erythroid transcription factor GATA1, and on distinctive alignment patterns that yielded a positive regulatory potential score. It was validated as a functional enhancer by both transient and stable reporter assays in human K562 erythroleukemia and murine erythroleukemia (MEL) cells, respectively.
