Pathways

p53 regulators

The list points to several regulators of p53 pathway. For more detailed information check the references PMIDs 16575405, 16601750, 19450511, 19410540. The genes are listed by type of action; each gene is listed with aliases in parentheses, full name and link to gene report pages:

Phosphorylation

Atm (ATA, ATE, TEL1) – ATM serine/threonine kinase
Chek1 (Chk1) – checkpoint kinase 1
Chek2 (Chk2) – checkpoint kinase 2
Note that Atm also phosphorylates Mdm2 of the negative feedback loop

Acetylation

Crebbp (CBP/p300, p300/CBP, CBP) – CREB binding protein (co-activator)
Kat5 (Tip60, Htatip1) – lysine acetyltransferase 5 (co-factor)
Kat8 (Myst1, Mof, hMOF) – K(lysine) acetyltransferase 8 (co-factor)
Note that Crebbp also acetylates Mdm2 of the negative feedback loop

Deacetylation

Hdac1 (Hd1, Rpd3) – histone deacetylase 1
Sirt1 (sirtuin (silent mating type information regulation 2 homolog) 1 (S. cerevisiae)) – sirtuin 1

Ubiquitination

Mdm2 (ACTFS) – Mdm2 proto-oncogene – in diagram
Rchy1 (Pirh2, Chimp, Arnip, hARNIP) – ring finger and CHY zinc finger domain containing 1
COP1 (RFWD2, Rnf200) – COP1, E3 ubiquitin ligase [human gene – the rat ortholog may not have enzymatic activity]
Note that these genes are also targets of p53

Deubiquitination

Usp7 (Hausp) – ubiquitin specific peptidase 7
Note that Usp7 also deubiquitinates Mdm2

Methylation

Setd7 (SET7/9, SET7, SET9) – SET domain containing (lysine methyltransferase) 7
Smyd2 (HSKM-B) – SET and MYND domain containing 2
