A pcDNA3 vector containing 2.1 ... (more) kb of human APP (isoform 770) cDNA was used to introduce the mutations Swedish K670N/M671L, Dutch E693Q, and Iowa D694N using the QuikChange kit. The APP770-SwDI cDNA was amplified by PCR using primers containing the NheI five linker and SacII three linker. The PCR product of the APP770-SwDI was subcloned between exons II and IV of a Thy1.2 expression cassette using NheI and SacII restriction sites. The completed construct was microinjected into pronuclei of Sprague-Dawley single-cell embryos. Founder rats were identified by Southern blot analysis of tail DNA.
